INVESTIGATION OF THE CYTOTOXIC EFFECT OF JUNIPERUS DRUPACEA ON GLIOBLASTOMA CELL LINE

Glioblastoma multiforme (GBM) is one of the most prevalent type of primary brain tumor with very poor prognosis. Juniperus drupacea is a popular medicinal plant which belongs to Cuppressaceae family. In this study investigation of the cytotoxic effect of Juniperus drupacea on glioblastoma cell line was aimed. For this purpose Juniperus drupacea cone seed extract was prepared and filter sterilized. The glioblastoma (GBM, U87 MG (ATCC® HTB-14™)) cell line was exposed to Juniperus drupacea extract, in the dilutions of 1/100, 1/80, 1/60, 1/50, 1/40, 1/20 and 1/10(v/v) for 1 and 3 days. Afterwards the viability of cells was evaluated by MTT cell viability assay. The viability assay showed that the increasing concentration of Juniperus drupacea extract given to cells caused the decreasing of cell viabilities. According to MTT results of 1 and 3 days ; 1/100, the lowest dilution of the Juniperus drupacea extract diminished the viability of cells by about 15% and %18 respectively, while 1/10, the highest dilution of the Juniperus drupacea extract diminished the viability of cells by about 65% and %74 respectively. The results revealed that Juniperus drupacea extract has a significant cytotoxic effect on U87MG cells. Thus it was considered that Juniperus drupacea active components could be potential medication candidates for the treatment of glioblastoma. But further investigations are needed.
